sounds to me like it might be a universal joint rather than the diff,
this is a slightly less costly opperation
about &163;7 per UJ, you need to get underneathe with the rear up on axel stands and wiggle each joint in the drive train to see which one has play in it, then remove as per haynes manual and fix it.

When the Diff is 'thinking' of expiring you'll hear either grinding noises from it or whining noises.
The single 'clunk' could be a number of things, but most likely a UJ (as mentioned), others could be loose flange bolts, worn mounting bushes, worn dampers, worn wheelbearings.
The latter of these will also have other symptoms eg fliud loss from damper, bouncy ride, wobbly wheel etc...

Sounds like something not properly tightened to me or it could be a UJ - although in my experience UJs tend to make a noise on over-run (prop shaft) or
going round bends (half shafts) rather than start up.
On the subject of filling, no, most Spitfire diffs do not have a drain plug and if the leak is "slight" it will take quite a long time before the level of oil in the diff drops sufficiently for any noticeable amount to go in. Do keep an eye on it tho - oil is cheaper than a diff.
